【原创】这篇文章从苜蓿中克隆得到了一个解旋酶-MH1,做了序列进化分析,亚细胞定位分析说明这个基因的特点,通过realtime PCR进行了四种非生物胁迫下的表达特性分析,发现全都明显上调表达,最后对拟南芥进行了超量表达,对转基因拟南芥做了抗逆性分析,发现提高了在干旱,盐,和氧化胁迫下的抗性,进一步分析机理,作者根据测定sod和apx活性发现上升,而钠离子并未发生明显变化推测MH1基因提高了抗逆性原因可能主要是提高清除ROS能力,从而提高对逆境的适应性,而不是常认为 的那样减少细胞内钠离子的浓度,这个推测比较有创新性,但就像作者最后说得那样,还需要进一步实验证明,另外,现在有很多人认为豆科来源的基因用模式植物拟南芥验证不一定准确,最好还能回归豆科植物来验证。 |
