本文介绍了三个光相关基因启动子的组织特异性,所采用的方法很简单,就构建promoter::GUS这样的植物表达载体,侵染拟南芥,观察表型。
本文值得学习的有两点:
1.数据来源与进化树的构建:Phylogenetic analysis of 73 A. thaliana PRXs was performed using the MEGA4 software package (Tamuraet al. 2007). Amino acid sequences of 73 A. thaliana PRXs and ZPO-C were aligned automatically using the CLUSTALW program (Thompson et al. 1994) included in MEGA4. Many gaps were observed in the initial part of aligned PRXs sequences; therefore, we manually excluded the initial sequences corresponding to positions 1–15 of AtPrx66 from the alignments. Aligned sequences were used to construct a phylogenetic tree using the neighbor-joining method (Saitou and Nei 1987). Evolutionary distances, which were computed using the Poisson correction method (Zuckerkandl and Pauling 1965), were expressed as the number of amino acid substitutions per site. All positions containing gaps and missing data were eliminated from the dataset (complete deletion option). There were 222 positions in the final dataset. Bootstrap values were obtained after 1000 replications.
2. 表型观测:作者对这三个转基因株系分别观察了幼苗、成苗期植株不同器官(根、茎、叶、花、种子)横切面及纵切面,具体鉴定其组织特异性,并做表显示(见表2,可惜没上传上来).
